The long filaments of microtubules are difficult to synthesize in a pure state. To circumvent this problem, we prepared a monodisperse population of microtubules by a one-step assembly of tubulin from guanosine triphosphate (GTP) in the presence of stabilizing agents and n-octylglucoside. As a result of polymerization, the polymer had a straight morphology, a uniform length, and a high monomer-to-polymer ratio. Because of the n-octylglucoside in the initial tubulin solution, depolymerization of the assembly occurred at a higher rate than the rate of assembly. We also measured the rate of depolymerization in bulk polymer solutions with and without exogenous GTP. In the absence of GTP, the depolymerization rate was about three times higher in the microtubules than in bulk. On the other hand, the addition of GTP increased the depolymerization
